The Ajanta Caverns are in the Aurangabad section of Maharashtra, Native indian. There are 29 rock-cut give ancient typical monuments which time frame from the 2nd millennium BC. New Fetal Genome Sequencing Could Spot Disorders

In a development scientists are calling a "tour de force," researchers have reconstructed the genome of a fetus using DNA samples from the parents.

Because their technique did not require an invasive test to take samples from the fetus itself, it's an important step toward what could become a low-risk way to identify genetic disorders early in development, experts say.

Currently, "when genetic testing is done, it's done for just a few diseases," said lead author Dr. Jay Shendure, an associate professor of genome sciences at the University of Washington.

A test based on the new technique could detect the roughly 3,000 conditions known as Mendelian disorders, each of which are the result of a single mutated gene, Shendure said. Huntington's disease, hemophilia and sickle-cell anemia fall into this category.

While each of these disorders is relatively rare, together they affect about 1 percent of births, Shendure said.

"This is amazing," said Dr. Ada Hamosh, director of the Institute of Genetic Medicine at the Johns Hopkins University School of Medicine, of the findings. "On the other hand, in no way is this ready for prime time," said Hamosh, who was not involved with the research.

Shendure and colleagues put together the fetal genome using a saliva sample from the father, and a sample of blood plasma from the mother. About 13 percent of the DNA found outside of cells in a pregnant woman's body belongs to her fetus.

They sequenced the regions of DNA they were aiming for with 98.2 percent accuracy.

Shendure estimated that reconstructing the fetal genome cost roughly between $ 20,000 and $ 50,000.

But with the cost of genome sequencing expected to come down, and as the technique is further refined, Shendure said, the hope is that researchers can develop a clinical test that detects many diseases noninvasively.

It's also hoped that testing could be done earlier in pregnancy than current tests, he said. In the study, the researchers sequenced the fetal genome twice, once in a fetus at 8 weeks into the pregnancy, and another time with a different fetus at 18 weeks. The highly accurate information only came at the later time.

The accuracy is affected variations between pregnancies, Shendure noted. "It's possible it could be done in the first trimester, it just may require more resources to do it."

At present, the only test routinely done that uses maternal blood plasma, as in the study, is for Down syndrome.

But to confirm the results of that blood test, an invasive test called amniocentesis, which requires fetal tissue samples, is used, said Dr. Laird Jackson, a professor of obstetrics and gynecology at the Drexel University School of Medicine.

Hamosh said that while a noninvasive test would be an improvement, at present, a full analysis of a fetus's genome would be overkill, because parents can be tested for specific genes of concern, and a fetus could be tested if the parents' tests reveal a potential problem.

"You don't need whole genome sequence information on a fetus," she said. If a family is known to carry a risky version of a gene, then a test for that gene can be done.

But as the techniques used in the study are improved, she said, they could be helpful in catching cases where new mutations arising in the fetus are the source of genetic diseases, rather than mutations inherited from the parents.

"I think this paper is a tour de force," Hamosh said. "This was unthinkable six months ago. Is it ready for any kind of clinical application? Not at all yet."

While a noninvasive test may be at least several years from the clinic, Jackson said, ultimately they could be a boon for parents, providing more of them with more information about their future children.

With current invasive tests, "if you go to the right person, if you go to a center that has excellent [health professionals], the risk of losing that pregnancy is so small," Jackson said.

But many mothers can't get to those top-notch facilities, he said.

A noninvasive diagnostic test could increase access, Jackson said. "Now mommy only has to go some place where someone can draw her blood."

The Ajanta Caves are in the Aurangabad district of Maharashtra, India. There are 29 rock-cut cave monuments which date from the 2nd century BC. The caves include paintings and sculptures considered to be masterpieces of both Buddhist religious art as well as frescos which are evocative of the Sigiriya paintings in Sri Lanka. Since 1983, the Ajanta Caves have been a UNESCO World Heritage Site. The caves are on a horse shoe shaped scarp. The most important and popular caves are the first, second and fourth. The first cave displays one of the most elaborate carvings on its facade with relief sculptures on entablature and ridges. There are scenes carved from the life of the Buddha as well as a number of decorative motifs. A two pillared portico, visible in the 19th-century photographs, has since perished. The cave has a front-court with cells fronted by pillared vestibules on either side. These have a high plinth level. The cave has a porch with simple cells on both ends.

The second cave is known for the paintings that have been preserved on its walls, ceilings, and pillars. The cave is supported by strong pillars, festooned with designs. The front porch consists of cells supported by pillared vestibules on both ends. The hall has four colonnades which are supporting the ceiling and there are exquisite paintings in the halls. The painted narratives of the Jataka tales are depicted only on the walls, which demanded the special attention of the devotee.

The fourth cave is an unfinished grand scale monastery. The unfinished monastery is architecturally a wonder and contains a massive Buddhist idol flanked by celestial nymphs and devotees.

Ellora is an archaeological site, 30 km from the city of Aurangabad in the Indian state of Maharashtra built by the Rashtrakuta dynasty . It is well-known for its enormous caves; Ellora is also a World Heritage Site. The thirty four caves are actually structures excavated out of the vertical face of the Charanandri hills. Buddhist, Hindu and Jain rock-! cut temp les were excavated here. There are two Buddhist caves. Cave 1 is a vihara with eight cells, four in the back wall and four in the right wall. It had a portico in the front with a cell. The Vishvakarma (Cave 10) is the only chaitya griha amongst the Buddhist group of caves. The Hindu caves were constructed between the middle of sixth century to the end of the eighth century. These structures represent an unusual style of creative vision and implementation skills. Some were of such intricacy that they required several generations of planning and co-ordination to complete.

The five Jain caves at Ellora belong to the ninth and tenth centuries. They reflect a strict sense of asceticism which is the essence of Jainism.

Stress Statistics - Does Work Related Stress Give you Anxiety?

All signs stress statistics taken recently, it happens to be found that work related stress will be the leading grounds for anxiety. The old Greek and Egyptian philosophers taught that no child came with a vacant mind. Each child comes with a hundred tendencies generated by past conscious actions. It did not acquire these in this life,and we are bound to admit that it must have had them in past lives. The rankest materialist has to admit that these tendencies are the result of past actions,only they add that these tendencies come through heredity. Our parents, grandparents, and great - grandparents come down to us through this law of heredity. Now if heredity alone explains this, there is no necessity of believing in the soul at all, because body explains everything.

We need not go into the different arguments and discussions on materialism and spiritualism. So far the way is clear for those who believe in an individual soul. We see that to come to a reasonable conclusion we must admit that we have had past lives. This is the belief of the great philosophers and sages of the past and of modern times. Such a doctrine was believed in among the Jews. Jesus Christ believed in it. He says in the Bible, "Before Abraham was, I am." And in another place it is said, "This is Elias who is said to have come."

All the different religions which grew among different nations under varying circumstances and conditions had their origin in Asia, and the Asiatics understand them well. When they came out from the motherland, they got mixed up with errors. The most profound and noble ideas of Christianity were never understood in Europe, because the ideas and images used by the writers of the Bible were foreign to it.Take for illustration the pictures of the Madonna. Every artist paints his Madonna according to his own pre-conceived ideas.

The lies of some modern day Zen masters

Some modern day Zen masters teach that in Buddhism there is no further existence or rebirth after death.  But if this is true why, for example, in some of the older discourses of the Buddha, the Buddha says that a "fool who has not lived the holy life for the complete destruction of suffering....with the breakup of the body, the fool fares on to another body" (S. ii. 24)? 

Besides this, it is said that the Buddha could remember his former existences  prior to becoming a Buddha.  Therefore, how might this ability to recall one's former existences be accomplished if death is just death without rebirth? Practically the whole point of Buddhism is to end the cycles of rebirth—again birth and again death.   

As regards Zen, if there is no life after death then why did Bodhidharma say the following?

"The Buddha said people are deluded. This is why when they act they fall into the River of Endless Rebirth. And when they try to get out, they only sink deeper. And all because they don't see their nature."

I haven't a clue as to why some Zen masters peddle this inaccurate view of Buddhism that there is no re-existence after death.  Is it just to attract Western followers who believe death is final?  It almost goes without saying that the Western view of death is, bluntly, brutish.  

The power which organizes the body and keeps its from disintegrating is greater than the sum of the body's anatomical parts.  This passage from the Buddha in the Maharatnakuta Sutra sheds some light on this.

"From consciousness the body arises, and consciousness covers all the body and limbs.  When we look for consciousness in the body, we cannot find it anywhere; yet without consciousness, the body cannot live." 

In this Sutra, consciousness (citta/vijnana) is prior to the corporeal body in which the body is being, figuratively speaking, is submerged in the pure medium of consciousness.  Given that consciousness is always prior, the body is ever posterior always being subject to death.  Thus, when death comes to the body, it doesn't reach as far as consciousness.  Again, citing from the Maharatnakuta Sutra, the Buddha says:

"Just as a silkworm makes a cocoon in which to wrap itself and then leaves the cocoon behind, so consciousness produces a body to envelop itself and then leaves that body to undergo other karmic results in a new body."

According to the Buddha, we must keep in mind that our temporal body, which is made up of biological tissue, "is not yours, nor does it belong to others.  It should be regarded as previous karma effected through what has has been willed and felt" (S. ii. 65).  In his regard, it is implied that we are, as consciousness, fundamentally deathless.  It is only when consciousness finds support in biological tissue "is the genesis of the whole mass of suffering" (S. ii. 65).  Were consciousness not to glom into such embryonic tissue, for example, there would not be the mass of suffering.  Nor would there follow old age and death.

Onward goes the idea of the material world, onward moves the idea of freedom. Still the fight goes on. We are hearing about all the quarrels of creeds and sects,yet creeds and sects are just and proper,they must be there. The chain is lengthening and naturally the struggle increases, but there need be no quarrels if we only knew that we are all striving to reach the same goal.

The embodiment of freedom,the Master of nature, is what we call God. You cannot deny Him. No, because you cannot move or live without the idea of freedom. Would you come here if you did not believe you were free? It is quite possible that the biologist can and will give some explanation of this perpetual effort to be free. Take all that for granted, still the idea of freedom is there. It is a fact, as much so as the other fact that you cannot apparently get over, the fact of being under nature.

Bondage and liberty, light and shadow, good and evil must be there, but the very fact of the bondage shows also this freedom hidden there. If one is a fact, the other is equally a fact. There must be this idea of freedom. While now we cannot see that this idea of bondage, in uncultivated man, is his struggle for freedom, yet the idea of freedom is there. The bondage of sin and impurity in the uncultivated savage is to his consciousness very small,for his nature is only a little higher than the animal's. What he struggles against is the bondage of physical nature, the lack of physical gratification, but out of this lower consciousness grows and broadens the higher conception of a mental or moral bondage and a longing for spiritual freedom. Here we ! see the divine dimly shining through the veil of ignorance. The veil is very dense at first and the light may be almost obscured,but it is there,ever pure and undimmed -- the radiant fire of freedom and perfection. Man personifies this as the Ruler of the Universe, the One Free Being. He does not yet know that the universe is all one, that the difference is only in degree, in the concept.

Yoga Class Review - My Lovely Lovely Hips

So here it goes. I'm starting a blog to review on-line yoga classes. I have to say that initially I didn't think the idea of on-line classes made sense. I've always loved the feeling of being in a class surrounded by many people. But now I get it. I no longer live in close proximity to the classes I want to attend and I don't have the luxury of spending 2+ hours on my yoga classes anymore (I used to walk for an hour to my class in London and then stroll back home afterwards to avoid the public transport option!). So when faced with the choice of: a) no yoga / not so great yoga, or b) excellent yoga taught by accomplished teachers on-line whenever you want to do it, it all started to make sense!

So the first class I checked out is on My Yoga Online. Now I've heard this is the best of the on-line yoga websites but we'll see as I continue to review classes. I was really impressed with My Yoga Online - the process for signing up was easy and straightforward. You can either pay monthly / annually to view as many classes as you want or purchase a single class that you own (I bought this one for .99). Searching for classes is easy and straightforward and when I had an issue with streaming a video I got an answer back from customer service within 24 hours.

My Lovely Lovely Hips is a 55 minute class taught by Clara Roberts-Oss that is listed as Advanced. The filming was great - it was enjoyable to watch and Clara's voice and instruction were excellent. Some of the hip openers were pretty sophisticated so I did end up watching at times (as opposed to just listening to the instruction which is what I prefer to do normally in a class). The class was easy to follow and I would agree that it is Advanced (or maybe somewhere between Intermediate and Advanced. Definitely not yoga for beginners). Clara starts off with some sun salutations and incorporates the hip openers nicely into the flow. She then moves into some backbends and forward bends before Sivasana. She even throws in a couple of balances ! (one inc orporating a hip opener). This really is a great class if you want to open up the hips - I'm very flexible so rarely find classes where the teachers go deep enough in terms of the stretch but this class did the trick. My only complaint is a personal pet peeve (so some will take this as a positive comment) which is that Clara jokes around a few times throughout the class (what I will call the Silliness Factor). I know some people enjoy classes where the teacher makes funny sounds or acts goofy but I do not, so just take it as you will.

On-Line Yoga Class Review - Royal Protocol Sun Salutations

Here is another review from My Yoga Online. At some point I will be reviewing classes from the other on-line yoga websites and will compare the different providers. But I'm going to do a few more My Yoga Online classes first because there are so many options in terms of teachers, styles of yoga, levels and even styles of classes (ie. classes filmed in the My Yoga Online studio like My Lovely Lovely Hips, instruction like Royal Protocol, live yoga classes filmed with students in a yoga studio).

Royal Protocol Sun Salutations is an instructional video on how to do a sun salutation. Royal Protocol is a seven minute class taught by Nico Luce and is listed as being appropriate for All Levels. I would say this is for beginners, however, for experienced practitioners it's always good to have a refresher of the basics - and it's a real treat just watching Nico's alignment and grace as he jumps back with oh so much control!

In this class Nico is speaking as he demonstrates the poses - in the previous two classes I reviewed (My Lovely Lovely Hips and Enliven Yourself) the teachers just demonstrated and their voices were recorded separately. I found that it made sense for Nico to speak as he was demonstrating since this is an instructional class and it seemed natural for him to speak as he moved through the poses. The filming is great with the camera moving to different angles so you can get a good look at the alignment for each pose.

I love the beginner instruction because Nico gives a sun salutation variation that almost anyone can do. Many people give up on yoga because they can't do a catturanga or can't fully straighten their legs to do a standing forward bend and get frustrated because most yoga classes have lots of sun salutations (and thus the catturangas and forward bends!). Nico addresses these issues by showing a sun salutation with bent knees on the ground (so no catturanga). For those with tight hamstrings Nico instructs you to keep the knees bent and gives good instruction ! to ensur e the back doesn't do the brunt of the work. So, no more excuses! This video gives a very simple and quick explanation of how to do a sun salutation that can be incorporated into any practice or put in place of other sun salutation variations taught in a class.
